Charles University, Faculty of Pharmacy in Hradec Králové Department of Pharmacognosy and Pharmaceutical Botany Candidate: Ing. Rozálie Peřinová Supervisor: prof. Ing. Lucie Cahlíková, Ph.D. Title of doctoral thesis: Amaryllidaceae alkaloids of haemanthamine structural type and their semisynthetic derivatives as potential drugs in the treatment of Alzheimer's disease. To deepen the knowledge about the Amaryllidaceae alkaloid haemanthamine, which was isolated at our workplace as part of previous phytochemical studies, derivatives of this alkaloid were synthesized. First, series of aliphatic (3-12) and aromatic ester derivatives (13-66) were prepared, and then, to compare the structure-activity relationship, ether derivatives (67-80) were prepared from the most active substituents. All synthesized compounds were identified using the following structural analysis methods: NMR, HPLC/MS, and HRMS, including testing physical properties such as optical rotatability. After structure confirmation, all derivatives were subjected to screening studies for their inhibitory potential against hAChE and hBuChE. The selected derivatives were tested for their inhibitory potential against another enzyme, GSK-3β, which plays a significant role in the pathogenesis of AD. In cooperation with the Faculty of Medicine in...

Charles University, Faculty of Pharmacy in Hradec Králové Department of Pharmacognosy and Pharmaceutical Botany Candidate: Mgr. Rudolf Vrabec Supervisor: prof. RNDr. Lubomír Opletal, CSc. Title of doctoral thesis: Alkaloids from Vinca minor L. and their biological activity Key words: Vinca, indole alkaloids, Alzheimer's disease, AChE, BuChE, POP, GSK-3β, cytotoxicity, docking study Twenty-three monoterpene indole alkaloids of different structural types were isolated by chromatographic techniques from aerial parts of Vinca minor L. (Apocynaceae). The alkaloids were identified by a combination of analytical methods (NMR, MS, HRMS, optical rotation). Among obtained compounds, one structure was undescribed so far and was named vincaminorudeine. Eleven alkaloids were isolated for the first time from this species. Obtained compounds that were isolated in sufficient amounts were subjected to in vitro tests for the inhibition of hAChE, hBuChE, POP, and GSK-3β - enzymes that play a key role in the pathophysiology of Alzheimer's disease. The most active alkaloid was (−)-2-ethyl-3[2-(3-ethylpiperidinyl)-ethyl]-1H- indole (VR-19) with IC50 = 0,65 µM for the inhibition of hBuChE, and with IC50 = 58 µM for the inhibition of POP. Other alkaloids which exhibited a significant inhibition against hBuChE (IC50  30...

Functional and emotional impulses during shopping at the point of sale
Šteflová, Aneta ; Jesenský, Daniel (advisor) ; Koudelková, Petra (referee)
The Bachelor's thesis entitled "Functional and emotional impulses during shopping at the point of sale" addresses the description of buying impulses in a retail store. It illustrates those marketing possibilities for application of impulses in communication of non-chocolate sweets at the point of sale in hypermarkets. The first theoretical part defines shopping at the point of sale, its terminology, provides information about impulsive shopping and analyse particular buying impulses and possibilities of their usage. In the second part of the theses, theoretical knowledge is used from the first part to describe in-store marketing communication tools employed by different brands of non- chocolate category at the point of sale of Czech hypermarkets. Information about buying impulses is gained from literature and on-line sources. Information about POP communication tools in hypermarkets comes from my own observation and from analytic discussion of impulses' usage with materials gathered from the theoretical part. The main focus is on description of the current situation in application of POP communication tools, especially buying impulses used in communication of sweets with customers at the point of sale.
Evolution of western popular music and mainstream musical taste 2010-2019
Severin, Ivan ; Kruml, Milan (advisor) ; Hroch, Miloš (referee)
1 Abstract This thesis aims to map the evolution of western contemporary music ranging from year 2010 to 2019 due to a lack of information about it's development compared to previous decades. The recurring theme in this thesis is our paradigm shift into the age of new digital media. First chapter unwraps all the changes related to a move from old physical means of music distribution into the new world of digital platforms such as Spotify or Apple Music. I set out to show just how much this fact affected all parts of music creation all the way to its consumption. What follows then is an attempt to answer the question if today's music is in fact of a lower or higher quality than music of past decades. Next chapter describes the new phenomenon of so called supercomposers, the kind of composers that affected the sound of music we hear everyday with their handwriting. Actual production techniques and ways to make music are the next topic of this work alongside the explanation of how accessible these tools are to a new generation of musicians. Alongside to changes new media brought to the music world I set to explore the ways modern musicians use social media to gain their following and expand their audience. To close out the thesis I present several examples of how specific genres came to be affected by the rise...
Patterns of use addictive substances at active musicians in music groups in the Benešovsko
Olišarová, Pavlína ; Šejvl, Jaroslav (advisor) ; Kulhánek, Adam (referee)
Introduction: Addictive substances among musicians is a common occurrence. However, this phenomenon is mostly described only by the musicians themselves in their autobiographies. There is little professional work to address this phenomenon. There are many factors that affect musicians and lead them to use addictive substances. Even though this phenomenon is so widespread, it is taken for granted and few pay attention to it. Aim: Aim of thesis is to identify and describe patterns of substance use in active musicians in music groups that are based in the Benešov region, but play outside this area too. Methodology Data were collected in the form of a semi-structured interview. A list of music groups was created using the bandzone website. These groups were then found on Facebook to find out their activity. They were also subsequently approached here. 9 researched musicians took part in the research. Responses from the interviews were recorded directly by hand. Overwrite was performed while data was being written. Subsequently, the data were analyzed by elements of grounded theory. Using open coding, the codes were assigned to important topics in the text. These topics were grouped using axial coding. Using these categories, patterns of substance use among active musicians in music groups based in the...
